So I'm writing a book about two characters who are based off of people in my real life. They won't give me permission to use their names. If I change their names ( i.e- James Russel to Jack Roland) but keep their stories, physical description, ect., can I be sued for slander? I would really appreciate your feedback.

Changing names does not protect you from a law suit. The people in question could sue you regardless of what steps you take to insulate yourself. The real question is whether such a claim would hold up in court.  The answer to that is - it depends on the facts. By the way, written defamation is Libel, not Slander.  Check out some basic information on Wikipedia.  If you are still concerned, talk to you publisher or editor.
